Expression of Human Blood Coagulation Factor ¥· in Chinese Hamster Ovary Cells

Abstract

|
The synthesis and secretion of a biologically active human blood coagulation factor ¥· (F¥·) in eukaryotic cell culture system has been designed. B-domain deleted F¥· cDNA inserted into pCMV¥â plasmid was cotransfected with pRSVneo into Chinese hamster ovary (CHO) cells for the production of F¥· protein. The expression of B-domain deleted F¥· mRNA was detected by reverse transcriptase polymerase chain reaction (RT-RCR), and the presence of a biologically active F¥· protein in the culture media of cells was confirmed. These results indicate that the expression of F¥· cDNA in eukaryotic cells may be applicable for the production of biologically active protein.
